I haven't read Dorohedoro manga yet, But I got feeling that Nikaido past is the most painful from your list

  I don't know her backstory fully, but they foreshadowed it and it seems like it was terrible.
  Now that I look at my favorites none really have painful backstories

  Kinda.... his family died due to a disease, and his friend took the women he was gonna marry. He was also broke and had to join the military for food and shelter.
